Sabemos que estudi\u00f3 en la Universidad de Val\u00e8ncia, ampliando sus estudios despu\u00e9s en Sevilla y Madrid, donde cambi\u00f3 su nombre, Alfonso, por Diego. Fue escogido para acompa\u00f1ar a los hermanos Garc\u00eda Nodal en la expedici\u00f3n que en 1618 march\u00f3 al estrecho de Magallanes buscando seguridad para las flotas que iban de Espa\u00f1a a Filipinas. En este viaje reconoci\u00f3 el estrecho, denominando isla de X\u00e0tiva en la actual Tierra de Fuego, cambio impuesto por Felipe V. Descubri\u00f3 otro estrecho, el de San Vicente \u2013hoy canal Beagle\u2013, haciendo una descripci\u00f3n de sus puntas, dando a una de ellas el nombre de Cabo Setabense, y al grupo de islas m\u00e1s meridional del continente americano, Diego Ram\u00edrez. Finalizado su viaje escribi\u00f3 la obra: Reconocimiento de los estrechos de Magallanes y San Vicente y algunas cosas curiosas de navegaci\u00f3n (1621), manuscrito existente en la Biblioteca Nacional, que contiene observaciones sobre mareas, corrientes, m\u00e9todos de navegaci\u00f3n, etc. de gran importancia en su \u00e9poca. Por sus m\u00e9ritos fue nombrado por el rey Felipe III cosm\u00f3grafo real y piloto mayor de la Casa de Contrataci\u00f3n (1620).
